The individual users should not work on local databases. They should work on a database on the server, and (if/when they need it) use a local replica.
The .NSF of the server has a property where it says what template (.NTF file) it inherit it's design from. You don't need to type that in yourself, when you perform a Design Replace of a database, this will be populated from the template.
So the correct way to deploy an updated version of a database is to refresh the design from the template, and the users have access to the new version on teh server. The changes will be distributed to the local replicas at the next replication (assuming teh replication settings have been setup correctly, to include design changes).
The reason you work in a template is so you can apply the changes to a test database on a test server, test it and then deploy the new template into production.
